[Or live streamed via Zoom – sign up to receive link] \n\n\n\nRegistration\n\n\n\nBiography\n\n\n\n \n\n\n\nStefan Hell is a director at both the Max Planck Institute for Multidisciplinary Sciences in Göttingen and the Max Planck Institute for Medical Research in Heidelberg\, Germany. \n\n\n\nHe is credited with having conceived\, validated and applied the first viable concept for overcoming Abbe’s diffraction-limited resolution barrier in a light-focusing fluorescence microscope. For this accomplishment he has received numerous awards\, including the 2014 Kavli Prize in Nanoscience and the Nobel Prize in Chemistry. \n\n\n\nStefan Hell received his doctorate (1990) in physics from the University of Heidelberg. From 1991 to 1993 he worked at the European Molecular Biology Laboratory\, followed by stays as a senior researcher at the University of Turku\, Finland\, between 1993 and 1996\, and as a visiting scientist at the University of Oxford\, England\, in 1994. In 1997 he was appointed to the MPI for Biophysical Chemistry (named Max Planck Institute for Multidisciplinary Sciences since 2022) in Göttingen as a group leader\, and was promoted to director in 2002. From 2003 to 2017 he also led a research group at the German Cancer Research Center (DKFZ). SUMMARY:Phase separation in cell physiology and disease
DESCRIPTION:Speaker: Anthony Hyman\, Director General EMBLHost: Jan Ellenberg\, Director SciLifeLab \n\n\n\nCells organize many of their biochemical reactions by formation and dissolution of non-membrane-bound compartments. Recent experiments show that one common mechanism for such biochemical organization is phase separation of disordered proteins to form compartments with liquid properties. \n\n\n\nThese compartments can subsequently harden to form compartments with new material properties such as gels and glasses. These compartments can be described by principles elucidated from condensed-matter physics and are therefore termed biomolecular condensates. I will discuss potential roles of condensates in organization and robustness of cellular biochemistry and how they fail in neurodegenerative disease\, using aggregation of TDP-43 in ALS as a model. \n\n\n\nregistration\n\n\n\nBio \n\n\n\nThe Hyman lab studies how phase separation impacts the formation of membraneless compartmentalisation of macromolecules inside living cells. Stemming from our work on C.elegans in 2009 (Brangwynne et al. 2009)\, observations over the last decade have shown that many non-membrane compartments have liquid-like properties (Banani et al. 2017). The liquid-like nature of condensates is ascertained using microscopy in cells by observing fusion events\, round shape\, rapid diffusion of components\, and a predictable response to changes in thermodynamic parameters such as temperature. Phase separation phenomena can be used to describe the formation of P granules (Brangwynne et al. 2009)\, nucleoli (Brangwynne et al. 2011)\, stress granules and centrosomes (Brangwynne et al. 2009) and estimates suggest that at least 30% of proteins in the nucleus are in such compartments (Thul et al. 2017). SUMMARY:Cryo-electron microscopy as a discovery tool in sperm biology
DESCRIPTION:Speaker: Tzviya Zeev-Ben-Mordehai\, Utrecht University \n\n\n\nAbstract\n\n\n\nCryo-EM single-particle analysis (SPA) approaches typically entail overexpression and purification of the target complex. For that\, prior knowledge of the composition of the complex of interest is required. In the highly specialised sperm cell\, the molecular composition is often not known. During my talk\, I will show how we expand the range of samples attainable for SPA towards more native samples\, specifically towards complex macromolecular assemblies that cannot be reconstituted\, and show how SPA is used as a discovery tool in sperm biology. \n\n\n\nBiography\n\n\n\nAn expert in reproductive structural biology. Zeev-Ben-Mordehai received her PhD from the Weizmann Institute of Science in 2008. In 2015 she established her group in the Division of Structural Biology of Oxford University. Since 2017 her group is part of Bijvoet Centre for Biomolecular Research of Utrecht University. Her group developed the first cryo-ET workflow for mammalian sperm\, enabling molecularresolution imaging of flagellar ultrastructure across species. Her landmark studies have revealed the multi-scale architecture of mammalian sperm flagella (EMBO J 2021)\, structural specialisations of the sperm tail (Cell 2023)\, structural diversity of axonemes across mammalian motile cilia (Nature 2025)\, and in-cell structures at the mitochondria– cytoskeleton interface in sperm (PNAS 2021). DESCRIPTION:The Cell Biology at Scale (CB@S) meeting is co-hosted by SciLifeLab\, the national research infrastructure for life science in Sweden\, and Biohub. This event aims to bring together researchers working at the interface of cell biology\, imaging\, functional genomics\, computational biology\, and artificial intelligence. \n\n\n\nRecent advances in multiplexed molecular profiling\, high-resolution imaging\, perturbation technologies\, and AI-driven modeling are changing how we study cells. Conducting cell biology at scale is not simply about increasing throughput. It represents a shift in how experiments are designed\, how data are integrated\, and how biological questions are formulated across molecular\, cellular\, and organismal levels. \n\n\n\nCB@S 2026\, a one-day symposium\, will focus on forward-looking perspectives: \n\n\n\n\nHow do we integrate multimodal cellular data?\n\n\n\nHow can modeling and AI help build predictive\, cell-scale theoretical frameworks?\n\n\n\nWhat experimental and computational strategies are still missing?\n\n\n\n\nThe symposium will be accessible both on site and via live stream. \n\n\n\nThe annual CB@S meetings started four years ago\, and so far\, all have taken place in the United States. Now for the first time\, the meeting will be hosted in Europe. SUMMARY:Challenges in GPCR Drug Discovery
DESCRIPTION:G protein-coupled receptors (GPCRs) are the largest protein family encoded by the human genome and one of the key targets for therapeutic approaches across many disease areas. However\, only around 25% of the druggable GPCRs have been tapped into. This presents a significant opportunity to develop novel medicines with high therapeutic impact for the GPCR class in numerous indications. \n\n\n\nThe Challenges in GPCR drug discovery workshop is co-organized by Gothenburg University\, Uppsala University\, and AstraZeneca to explore the latest advancements in GPCR drug discovery research. Our aim is to bring together renowned international experts from industry\, biotech\, academia and service providers to facilitate communication between scientific groups and research areas within GPCR drug discovery community. The workshop focusses on lead-generation aspects such as\, computational approaches\, structural biology and pharmacology in early drug discovery of GPCRs. We believe this will lead to uncovering common challenges and finding new innovative approaches to drug discovery for GPCRs\, and further strengthen the GPCR community in the Nordics. \n\n\n\nThe workshop has a maximum of 90 attendants generating an informal atmosphere encouraging discussions\, networking and information exchange. Participants are encouraged to submit poster abstracts. Registration deadline is 27th of April. SUMMARY:Live-Cell Super-Resolution Microscopy: Technological Progress\, Hybrid Methodologies\, and Applications in Omics
DESCRIPTION:Liangyi Chen\, New Cornerstone Investigator\, Institute of Molecular Medicine\, National Biomedical Imaging Center\, Peking University\, China. \n\n\n\nHost: Jan Ellenberg\, SciLifeLab \n\n\n\nAbstract\n\n\n\nIn this talk\, I will present our decade-long efforts to advance state-of-the-art live-cell super-resolution microscopy. By introducing spatiotemporal Hessian regularization\, we developed Hessian Structured Illumination Microscopy (Hessian-SIM)\, which substantially reduces phototoxicity and enables real-time\, long-term imaging of dynamic structures such as mitochondrial cristae and insulin vesicle fusion pores in living cells. We next engineered a dual-modality fluorescence/label-free microscope for panoramic super-resolution imaging\, facilitating the discovery of novel organelles. Our 4Pi-SIM and 3D multiplane SIM (3D-MP-SIM) techniques deliver ~100-nm isotropic resolution while achieving a tenfold increase in volumetric imaging speed for live cells.  \n\n\n\nFurthermore\, we introduced a universal sparse deconvolution algorithm that overcomes the optical Rayleigh diffraction limit. When applied to structured illumination\, it enables 60-nm resolution at 564 Hz for sustained live-cell imaging and enhances resolution in multiphoton and light-sheet microscopy in vivo. Finally\, I will discuss our recent developments in a SIM-FLIM system and the integration of live-cell super-resolution SIM imaging with single-cell RNA sequencing to bridge nanoscale dynamics with transcriptomic profiles.

SUMMARY:Application of advanced imaging technology for cell-type specific neural\, neuromodulatory and astrocytic signalling in vivo
DESCRIPTION:Sofie Ährlund-Richter\, The Picower Institute for Learning and Memory\, Massachusetts Institute of Technology (MIT) \n\n\n\nTitle: Prefrontal Cortex Feedback Reshapes the Representation and Routing of Visual Information \n\n\n\nHow does the brain determine which sensory signals are behaviorally relevant\, and how are these signals routed across cortical networks? Top-down feedback from the prefrontal cortex (PFC) has long been proposed to play a central role in shaping sensory processing\, yet the circuit-level mechanisms by which distinct prefrontal pathways modulate sensory representations remain poorly understood. \n\n\n\nHere we investigate how anatomically and functionally distinct prefrontal subregions influence visual processing in primary visual cortex (VISp). Using a combination of axonal tracing\, two- photon calcium imaging\, and projection-specific chemogenetic perturbations in behaving mice\, we show that inputs from the anterior cingulate cortex (ACA) and orbitofrontal cortex (ORB) exert dissociable and context-dependent effects on visual responses. Our findings demonstrate that prefrontal feedback is not a uniform gain signal but instead operates through modular pathways that differentially shape sensory representations according to internal state and behavioral context. Building on these results\, we are developing next-generation in vivo approaches to uncover the cell-type-specific logic underlying cortical communication. By combining spectral unmixing–based two-photon imaging\, projection-defined labeling\, and targeted perturbations\, we can simultaneously image multiple\, defined VISp neuron populations distinguished by their long-range outputs. This strategy enables direct observation of how prefrontal feedback redistributes visual information across parallel cortical output channels in real time. \n\n\n\nTogether\, this work reveals how top-down control dynamically restructures sensory representations and routing in the cortex. By defining circuit principles of prefrontal modulation\, these findings provide a framework for understanding how disruptions in feedback control may contribute to altered perception and cognition in neurodevelopmental disorders. \n\n\n\nTalk 2:  \n\n\n\nIakovos Lazaridis\, McGovern Institute for Brain Research\, Massachusetts Institute of Technology (MIT) \n\n\n\nTitle: Striosomes and astrocytes shape striatal dopamine during behavioral state transitions \n\n\n\nAbstract: Basal ganglia circuits\, modulated by striatal dopamine\, support reinforcement learning\, action selection\, and behavioral flexibility. Integrating two independent lines of evidence\, we propose a framework in which striosomes set nigrostriatal output while dorsal striatal astrocytes gate local dopamine availability during behavioral state transitions. Using projection-defined circuit mapping and perturbations\, we show that striosomal D1 spiny projection neurons (SPNs) project directly to the substantia nigra pars compacta (SNpc)\, whereas striosomal D2 SPNs route indirectly via a central external globus pallidus node\, forming opponent pathways whose net influence on dopamine differs from matrix circuits. Astrocytes exhibit Ca²⁺ transients time-locked to SNpc-evoked dopamine release\, and selective astrocyte activation reduces subsequent dopamine availability without affecting spontaneous locomotion. In a probabilistic decision task\, large astrocytic Ca²⁺ events are enriched during disengagement and precede re-engagement; astrocyte signals predict upcoming state transitions and engagement more strongly than trial outcomes\, consistent with state-level regulation rather than action coding. Dopamine-dependent astrocytic adenosine release may provide a local mechanism for regulating dopamine availability while biasing D1/D2 signaling\, enabling flexible rebalancing of striatal output across behavioral states. SUMMARY:Decoding Enhancer-Mediated Gene Regulation with CAGE-seq and Deep Learning
DESCRIPTION:Michiel de Hoon\, RIKEN center\, Japan \n\n\n\n\n\n\n\nAbstract\n\n\n\nPrecise regulation of gene expression is fundamental to cellular identity and behavior\, and gene dysregulation is a common feature across a wide variety of diseases.  At the molecular level\, this regulation is orchestrated by transcription factors acting on non-coding regulatory elements at promoters and distal enhancers. Genome-wide association studies have shown that disease-associated variants are enriched in enhancer elements\, and somatic mutations in these regulatory regions can contribute to diseases such as cancer. As it is often unclear how such variants influence gene expression\, connecting enhancers to their target promoters is a key step toward understanding the regulatory basis of pathogenic gene expression.Cap Analysis Gene Expression (CAGE-seq)\, originally developed at RIKEN\, provides an unbiased and quantitative genome-wide view of transcription initiation at single-nucleotide resolution. Beyond quantitating mRNAs and long non-coding RNAs\, CAGE-seq revealed widespread transcription at enhancers\, enabling their systematic identification and quantitative characterization. While traditional bioinformatics approaches have focused on genome annotation and expression analysis\, emerging neural network frameworks now enable inference of enhancer–promoter interactions directly from sequencing data. \n\n\n\nIn this presentation\, I will highlight the use of AI-based sequence-to-function frameworks that can predict transcriptional activity directly from genomic sequence. When combined with CAGE-seq\, these approaches enable the construction of context-specific regulatory maps linking enhancer activity to gene expression. Using examples from leukemia\, I will illustrate how CAGE-seq data\, analyzed using deep learning\, support a systematic interpretation of inter-patient heterogeneity in gene regulation. \n\n\n\nBiography\n\n\n\nI entered computational biology during my Ph.D. in Physics at the University of California\, Berkeley. After postdoctoral training in bioinformatics at the University of Tokyo and Columbia University\, I joined RIKEN in Yokohama\, where I became a core contributor to the FANTOM consortium\, an international effort to functionally annotate the mammalian genome. \n\n\n\nBetween 2014 and 2021\, I led the computational biology program of FANTOM6\, coordinating my research group and consortium-wide efforts to functionally annotate non-coding RNA. I am currently a tenured Senior Research Scientist at the RIKEN Center for Integrative Medical Sciences\, investigating gene regulatory mechanisms in disease\, particularly cancer. \n\n\n\nMore recently\, my research has focused on developing and applying AI-based sequence-to-function models to infer long-range gene regulatory interactions from genome and transcriptome data\, with the aim of understanding the regulatory basis of inter-patient heterogeneity in disease. \n\n\n\nHost: Pelin Sahlen pelin.akan@scilifelab.se

DESCRIPTION:Spotlight seminar series proudly welcomes Professor Vile R. I. Kaila to give a seminar at SciLifeLab Campus Solna. \n\n\n\n\n\n\n\nBiography\n\n\n\nVille R. I. Kaila received his PhD in biochemistry in 2009 from University of Helsinki. After a postdoc at the US National Institutes of Health (NIH)\, he was appointed a professorship at the Technical University of Munich in 2013. In 2019\, Kaila moved with his research group to Stockholm\, where he is currently professor at the Department of Biochemistry and Biophysics at Stockholm University and a Wallenberg Scholar. His research focuses on elucidating mechanistic principles of bioenergetic enzyme complexes by development of integrative computational and experimental approaches. Read more at http://villekaila.com/ \n\n\n\nAbstract\n\n\n\nBiological energy conversion is catalyzed by intricate protein complexes that transduce redox energy into proton currents across a biological membrane\, powering the cellular energy metabolism. Yet\, despite significant advances in recent years\, the molecular principles of these reactions remain poorly understood and pose major scientific challenges. In this talk\, I describe our integrative mechanistic exploration of complex bioenergetic machines\, where we combine multiscale simulations and data-driven methods with biophysical experiments and structural (cryo-EM) studies to derive a molecular understanding of intricate long-range charge transfer reactions in biology on a broad range of timescales and spatial resolutions. I describe our recent work on understanding enzymes catalyzing long-range charge transfer reaction by intrinsic electric field effects. We find that mutations of key residues mediating these allosteric processes lead to perturbed coupling effects\, and development of severe human diseases\, with striking similarities in different biological systems. \n\n\n\n \n\n\n\nHost: Marta Carroni marta.carroni@scilifelab.se

SUMMARY:Molecular Mechanism of RNA Splicing by the Spliceosome
DESCRIPTION:Seminar by Yigong ShiWestlake University\, Hangzhou\, Zhejiang 310024\, China \n\n\n\nHost: David Drew\, SciLifeLab \n\n\n\nSplicing of the precursor messenger RNA (pre-mRNA) is an indispensable step in the information flow from DNA to protein in all eukaryotes. Research of the past four decades has led to accumulation of a large body of biochemical and genetic information on pre-mRNA splicing. Structural information is central to the elucidation of the molecular mechanism of pre-mRNA splicing by the spliceosome. In 2015\, we reported the first atomic structure of an intact spliceosome from S. pombe at an average resolution of 3.6 Å. This structure reveals the overall organization and the conserved atomic features of an intact spliceosome\, which turns out to be a protein-orchestrated metalloribozyme. Since then\, atomic structures have been reported for nearly all major functional states of the spliceosome\, allowing recapitulation of the entire splicing cycle and mechanistic understanding of RNA splicing. In this presentation\, I will summarize these advances and discuss implications on disease treatment.

SUMMARY:[Spotlight seminar] Systems Human Immunology and AI: Immune Setpoint and Immune Health
DESCRIPTION:Speaker: Professor John Tsang\, Yale University \n\n\n\nAbstract\n\n\n\nThe immune system\, critical for human health and implicated in many diseases\, defends against pathogens\, monitors physiological stress\, and maintains tissue and organismal homeostasis. It exhibits substantial variability both within and across individuals and populations. Recent technological and conceptual progress in systems human immunology has provided predictive insights that link personal immune states to intervention responses and disease susceptibilities. Artificial intelligence (AI)\, particularly machine learning (ML)\, has emerged as a powerful tool for analyzing complex immune data sets\, revealing hidden patterns across biological scales\, and enabling predictive models for individualistic immune responses and potentially personalized interventions. Here I light our recent work in deciphering human immune variation and predicting outcomes\, particularly through the concepts of immune setpoint\, immune health\, and use of the immune system as a window for measuring health.  \n\n\n\nBiography\n\n\n\nJohn Tsang is a systems immunologist\, computational biologist\, and engineer. He is the Anthony N. Brady Professor of Immunobiology and Biomedical Engineering at Yale University\, the Founding Director of the Yale Center for Systems and Engineering Immunology (CSEI)\, the Yale Lead and an Investigator at the Chan Zuckerberg Biohub New York\, and the co-Chief Scientific Officer of the Human Immunome Project (a non-profit dedicated to mapping human immune variation around the globe to develop AI models of human immune health for the benefit of all). Tsang has pioneered and established the concepts and predictive metrics of human immune setpoints and immune health including responses to vaccines\, infections\, autoimmunity\, and therapeutics in human populations spanning the lifespan from infancy to old age. Tsang is also interested in programming immune cells to enable longitudinal immune and health monitoring of the entire body. Tsang has received numerous awards including multiple NIH Merit Awards for his research and leadership in systems immunology\, COVID-19\, and human immunology. He has advised numerous programs and organizations\, including the Allen Institute\, the Gates Foundation\, CEPI\, World Allergy Organization\, National Cancer Institute\, National Institute of Allergy and Infectious Diseases\, National Institute of Diabetes and Digestive and Kidney Diseases\, Snow Medical (Australia)\, and the Walter and Eliza Hall Institute (Australia). DESCRIPTION:Applications are now welcome for participation in this highly regarded and rewarding interactive workshop by Science and SciLifeLab. Designed for early career researchers (PhD students and postdocs) in SciLifeLab:s research community\, the workshop focuses on the publishing landscape\, paper preparation\, and the publication process. \n\n\n\n\n\n\n\nWorkshop leaders: \n\n\n\n\nValda Vinson (Executive Editor\, Science Magazine)\n\n\n\nSacha Vignieri (Deputy Editor Research\, Science Magazine)\n\n\n\n\nThe workshop is split into a hybrid morning session\, and an afternoon on-site workshop (see Curriculum for details). Questions from the participants will be addressed by the Editors in generous Q&A sessions.  \n\n\n\nOn-site participation\, including lunch and coffee breaks with the Editors\, is limited to 40 participants.
