SUMMARY:[Spotlight seminar] Systems Human Immunology and AI: Immune Setpoint and Immune Health
DESCRIPTION:Speaker: Professor John Tsang\, Yale University \n\n\n\nAbstract\n\n\n\nThe immune system\, critical for human health and implicated in many diseases\, defends against pathogens\, monitors physiological stress\, and maintains tissue and organismal homeostasis. It exhibits substantial variability both within and across individuals and populations. Recent technological and conceptual progress in systems human immunology has provided predictive insights that link personal immune states to intervention responses and disease susceptibilities. Artificial intelligence (AI)\, particularly machine learning (ML)\, has emerged as a powerful tool for analyzing complex immune data sets\, revealing hidden patterns across biological scales\, and enabling predictive models for individualistic immune responses and potentially personalized interventions. Here I light our recent work in deciphering human immune variation and predicting outcomes\, particularly through the concepts of immune setpoint\, immune health\, and use of the immune system as a window for measuring health.  \n\n\n\nBiography\n\n\n\nJohn Tsang is a systems immunologist\, computational biologist\, and engineer. He is the Anthony N. Brady Professor of Immunobiology and Biomedical Engineering at Yale University\, the Founding Director of the Yale Center for Systems and Engineering Immunology (CSEI)\, the Yale Lead and an Investigator at the Chan Zuckerberg Biohub New York\, and the co-Chief Scientific Officer of the Human Immunome Project (a non-profit dedicated to mapping human immune variation around the globe to develop AI models of human immune health for the benefit of all). Tsang has pioneered and established the concepts and predictive metrics of human immune setpoints and immune health including responses to vaccines\, infections\, autoimmunity\, and therapeutics in human populations spanning the lifespan from infancy to old age. Tsang is also interested in programming immune cells to enable longitudinal immune and health monitoring of the entire body. Tsang has received numerous awards including multiple NIH Merit Awards for his research and leadership in systems immunology\, COVID-19\, and human immunology. He has advised numerous programs and organizations\, including the Allen Institute\, the Gates Foundation\, CEPI\, World Allergy Organization\, National Cancer Institute\, National Institute of Allergy and Infectious Diseases\, National Institute of Diabetes and Digestive and Kidney Diseases\, Snow Medical (Australia)\, and the Walter and Eliza Hall Institute (Australia). Tsang earned his Ph.D. in biophysics from Harvard University and trained in computer engineering (BASc) and computer science (MMath) at the University of Waterloo\, Canada.  \n\n\n\nHost: burcu.ayoglu@scilifelab.se

Designed for early career researchers (PhD students and postdocs) in SciLifeLab:s research community\, the workshop focuses on the publishing landscape\, paper preparation\, and the publication process. \n\n\n\n\n\n\n\nWorkshop leaders: \n\n\n\n\nValda Vinson (Executive Editor\, Science Magazine)\n\n\n\nSacha Vignieri (Deputy Editor Research\, Science Magazine)\n\n\n\n\nThe workshop is split into a hybrid morning session\, and an afternoon on-site workshop (see Curriculum for details). Questions from the participants will be addressed by the Editors in generous Q&A sessions.  \n\n\n\nOn-site participation\, including lunch and coffee breaks with the Editors\, is limited to 40 participants. SUMMARY:[Spotlight seminar] - Designing brighter dyes for advanced fluorescence microscopy and beyond
DESCRIPTION:Dr. Luke D. Lavis\, Howard Hughes Medical Institute\, USA  \n\n\n\nBiography\n\n\n\n Luke D. Lavis received his B.S. in Chemistry from Oregon State University in 2000 and his Ph.D. in Organic Chemistry from the University of Wisconsin–Madison in 2008. Circumventing a traditional postdoc\, he than began his independent career as a Group Leader at the Howard Hughes Medical Institute’s Janelia Research Campus\, and was promoted to Senior Group Leader and Head of Molecular Tools and Imaging in 2017. His research focuses on designing small-molecule tools for imaging\, leading to the development of the widely used Janelia Fluor® dyes and the founding of Eikon Therapeutics in 2019.  \n\n\n\nAbstract\n\n\n\nSpecific labeling of biomolecules with bright\, photostable fluorophores is the keystone of fluorescence microscopy. An established method to label cellular components utilizes genetically encoded self-labeling tags\, which enable the attachment of chemical fluorophores to specific proteins inside living cells. For the last decade\, our laboratory has focused on the development of improved fluorophores for use in living systems. We discovered a simple structural modification to classic fluorophores—incorporation of a four-membered azetidine ring—that substantially improves quantum efficiency and photostability. This strategy is generalizable to fluorophores from different structural classes and has yielded a palette of optimized chemical dyes for use in advanced microscopy experiments ranging from single-molecule tracking in individual cells to population voltage imaging in vivo. \n\n\n\nHost: Simon Elsässer\, KI\, simon.elsasser@scilifelab.se

SUMMARY:How protein degraders work: Molecular mechanism and drug design
DESCRIPTION:Alessio Ciulli \n\n\n\n\n\n\n\nProfessor of Chemical Structural Biology \n\n\n\nFounder and Director\, Centre for Targeted Protein Degradation \n\n\n\nSchool of Life Sciences\, University of Dundee \n\n\n\nAbstract\n\n\n\nProtein degraders (also known as PROTACs and molecular glues) are a new class of small molecule tools and drugs that trigger the rapid depletion of a target protein inside the cell. Degraders work via forming a ternary complex\, bridging a target protein to a ubiquitin E3 ligase. This allows the E3 ligase to label the target protein with ubiquitin molecules\, leading to the protein being recognized and degraded by the proteasome. Whereas conventional drugs only inhibit disease-causing proteins by binding and locking up their most important functional parts for the duration of the drug’s action\, degraders can bind at any positions and cause the protein rapid and long-lasting destruction. This lecture will recount some of our fundamental discoveries and insights that have helped to advance understanding and illuminate the molecular mechanism of how degraders work. It will illustrate how such fundamental principles have been applied to design high-quality degraders against challenging drug targets\, and will highlight some of the latest progress from the laboratory. \n\n\n\nBiography\n\n\n\nAlessio Ciulli studied chemistry in Florence\, Italy\, and obtained his PhD from the University of Cambridge\, UK\, in 2006. After postdoctoral research in Cambridge and a brief visit at Yale University in the USA\, he returned to Cambridge in 2009 to start his independent laboratory. In 2013\, he moved to the University of Dundee\, UK\, where he was promoted to full professor in 2016. He has received numerous prizes and awards for his discoveries\, most recently the 2022 Prous Institute – Overton and Meyer Award for New Technologies in Drug Discovery from the European Federation of Medicinal Chemistry (EFMC). In 2023 he was elected Fellow of the Royal Society of Edinburgh. He is the scientific founder of Amphista therapeutics\, a targeted protein degradation company spin out of his laboratory\, and the founder and director of the University of Dundee’s new Centre for Targeted Protein Degradation which opened in January 2023. The Ciulli Laboratory focuses on chemical biology and structural biology to develop small molecules inducing targeted protein degradation and modulating protein-protein interactions and translates their fundamental research via collaboration partnership with the biopharma industry and by creation of spin-out companies.

Människan har cirka 20 000 gener som kodar för proteiner\, och HPA hjälper till att identifiera olika kombinationer av proteiner som är avgörande för mänsklig biologi. Som exempel kan cancertumörceller målsökas på detta sätt. Dessa målsökande bindarproteinerna kan “beväpnas”\, t.ex. med cellgifter eller lämpliga radioisotoper som dödar tumörcellerna\, med minimal påverkan på frisk vävnad. \n\n\n\nKlinisk GenomikAnna Lyander\, SciLifeLab/Karolinska Institutet \n\n\n\nClinical Genomics är en infrastrukturplattform vid Scilifelab. Plattformen arbetar med avancerade genomikmetoder\, olika typer av sekvenseringsteknologier och dataanalysverktyg\, för att stötta forskning och sjukvård. SUMMARY:Open online lecture by Prof. Edward Holmes: Metagenomics at the human-animal interface
DESCRIPTION:The PLP (Pandemic Laboratory Preparedness) program\, started in 2021\, acts to meet society’s need for efficient use of resources\, training and education\, to propagate skills and setting up technologies and equipment to be better prepared for new pandemics. \n\n\n\nDuring the upcoming PLP program retreat\, on Friday September 1 at 08:50\, we are honored to welcome Professor Edward Holmes for an online lecture\, open for everyone to join. \n\n\n\n\n\n\n\n\n\nEdward (Eddie) Holmes is an evolutionary biologist distinguished for his work on the emergence and evolution of viruses. He has used genomic and phylogenetic approaches to reveal the major mechanisms of virus evolution and determined the genetic and epidemiological processes that explain how viruses jump species boundaries and spread in new hosts.His work has revealed the origin\, evolution and molecular epidemiology of important human pathogens including influenza\, HIV and dengue\, and enabled more accurate assessments of what types of virus are most likely to emerge in human populations and whether they will evolve human-to-human transmission. His recent research has provided fundamental insights into the breadth and biodiversity of the viral world.Eddie is currently a Professor in the School of Medical Sciences at the University of Sydney. In 2015 he was elected a Fellow of the Australian Academy of Science and in 2017 he was elected a Fellow of the Royal Society. DESCRIPTION:Lunch seminar (also available through Streamo) where a new call for proof-of-concept funding aiming to ignite life science innovation will be presented.  \n\n\n\nRegister\n\n\n\nAre you financed through Knut and Alice Wallenberg Foundation\, a SciLifeLab researcher or infrastructure staff with ideas for commercialization\, this seminar is for you. \n\n\n\nLunch sandwiches will be served for participants attending in person. Register now on the link below. \n\n\n\nThe purpose of the Proof of Concept grant is to bridge the gap from academic research to innovations in life science. The grant gives researchers the opportunity to develop their early-stage discoveries towards validated methods\, products or processes and  provides the opportunity to carry out activities that validate and accelerate the development of the project and prepare for innovation and commercialization. \n\n\n\nThe Proof of Concept grant is not meant as a translational academic research grant\, but as a specific goal-oriented project to develop an academic discovery towards an innovation and a product. Hence\, translational research without a clear product-oriented thinking or a defined goal at the end of the two year period is not something that can be supported. The Proof of Concept grant is tied to the Wallenberg Launch Pad (WALP) mechanism (see below) that provides support for academic scientists to develop innovations that make an impact in society. \n\n\n\nThe approved funding for each project is suggested to be between 1 and 4 million SEK\, over a project period of a maximum of two years. The grant will be paid directly to the researcher’s university and the use of the grant must comply with the university’s regulations and the specific terms and conditions of the grant as stated by Knut and Alice Wallenberg Foundation (KAW). SUMMARY:SciLifeLab Group Leader Retreat
DESCRIPTION:We warmly welcome all SciLifeLab Group Leaders to take part in the  SciLifeLab’s Group Leader Retreat 2022\, June 14-15\, at the Aronsborg conference center in Bålsta. \n\n\n\nIn December 2019\, the SciLifeLab Board defined the criteria for being a SciLifeLab Group Leader. Currently\, there are close to 250 Group Leaders with a strong affiliation to SciLifeLab research and infrastructure. The Group Leaders are the core of the SciLifeLab community\, and are essential to the scientific development of our organization. \n\n\n\nThis two-day retreat is the first to bring together the expanding SciLifeLab Group Leader community from all SciLifeLab sites. The organizing committee (Anniina Vihervaara\, Sarahi Garcia\, Prashant Singh\, Jean Hausser\, Ellen Sherwood\, Mats Nilsson and Per Ljungdahl) has developed a program aimed at stimulating interactions and networking between Group Leaders\, catalyzed through activities such as speed dating\, research discussions\, and a sci-hack. Näär is a Professor of Metabolic Biology in the Department of Nutritional Sciences & Toxicology at the University of California\, Berkeley. He was a postdoctoral research fellow with Robert Tjian at UC Berkeley/HHMI\, where he discovered the human Mediator transcriptional co-activator complex. Dr. Näär was a Professor in the Department of Cell Biology\, Harvard Medical School and the Massachusetts General Hospital Cancer Center. A major focus of his lab is to understand transcriptional and microRNA regulatory mechanisms controlling metabolic homeostasis to guide novel therapeutic strategies for the treatment of cardiovascular disease\, obesity\, Type 2 diabetes\, non-alcoholic fatty liver diseases (NAFLD/NASH)\, age-related macular degeneration (AMD)\, Duchenne muscular dystrophy (DMD)\, multi-drug resistant fungal infections\, and numerous types of cancer. His work resulted in a number of impactful studies\, patents and foundation of companies.
